Proteins at the tRNA binding sites of Escherichia coli ribosomes.

چکیده
p - Nitrophenylcarbamyl - phenylalanyl-tRNA binds to ribosomes in response to poly(U) and reacts with ribosomal proteins via covalent bond formation. Ribosomal proteins of Escherichia coli were characterized by two-dimensional gel electrophoresis and by reaction with specific antibodies. The affinity label is shown to react with the 50S proteins L27, L15, L2, L16, and L14. We therefore conclude that these particular proteins are located at or near tRNA-binding sites within the 50S ribosomal subunit.

Function of the ribosomal E-site: a mutagenesis study
Ribosomes synthesize proteins according to the information encoded in mRNA. During this process, both the incoming amino acid and the nascent peptide are bound to tRNA molecules. Three binding sites for tRNA in the ribosome are known: the A-site for aminoacyl-tRNA, the P-site for peptidyl-tRNA and the E-site for the deacylated tRNA leaving the ribosome. Here, we present a study of Escherichia c...
متن کاملPhoto-affinity labeling of tRNA binding sites in macromolecules. I. Linking of the phenacyl-p-azide of 4-thiouridine in (Escherichia coli) valyl-tRNA to 16S RNA at the ribosomal P site.
The phenacyl-p-azide of 4-thiouridine in (E. coli) tRNA(1) (Val) was prepared for use as a photo-affinity probe of tRNA binding sites on ribosomes. The derivatized tRNA was 90-100% as active as control tRNA for aminoacylation, nonenzymatic binding to the ribosomal P site, elongation factor Tu(EFTu)-dependent binding to the A site, EFTu-GTP-aa-tRNA ternary complex formation, and transfer of vali...
متن کاملA photoaffinity labelling study of the messenger RNA-binding region of Escherichia coli ribosomes.
A photoaffinity labelling study of the messenger RNA-binding region of E. coli ribosomes has been made, using oligoadenylic acids as mRNA analogs. The oligonucleotides, of chain length 6 to 8 and thus several nucleotides longer than oligonucleotides previously employed for this purpose, carried a radioactive photolabile aromatic azide reagent bound covalently to the 3'-terminal ribose moiety. T...
متن کاملStructural requirements of Escherichia coli formylmethionyl transfer ribonucleic acid for ribosome binding and initiation of protein synthesis.
Escherichia coli formylmethionine tRNA can be isolated as two isomers which differ by a single nucleotide in the variable loop (m7G uersus adenosine). These two initiator tRNAs have been found to bind to ribosomes in an initiation factor and AUG-dependent reaction with identical kinetics. In addition, the bound tRNAs react with puromycin and initiate polypeptide synthesis at the same rates. Con...
متن کاملThe primary structure of protein L27 from the peptidyl-tRNA binding site of Escherichia coli ribosomes.
Bromoacetyl-phenylalanyl-tRNA(phe) bound to 70S E. coli ribosomes reacts covalently with proteins of the 50S subunit. The major reactions are with proteins L2 and L27. In the presence of poly(U), 70S-bound bromoacetyl-phenylalanyl-tRNA(phe) can participate in peptidebond formation with phenylalanyl-tRNA(phe) or puromycin. Most of the products of these reactions are also found covalently attache...
